Job Summary

A Research Technician II position is available in the lab of Dr. Christopher Mount in the MGH Pathology Department and Krantz Family Center for Cancer Research. This position requires an individual who will work independently under supervision to generate and evaluate cellular immunotherapies in brain tumor model systems. With training and supervision, this individual will also perform molecular characterization assays including single cell sequencing, spatial transcriptomics, and multiplexed immunofluorescence. This position will also require entry-level laboratory management, including monitoring of inventory, package receiving and shipping, routine instrument maintenance, and interfacing with core facilities and collaborators to facilitate specimen handling.

The lab develops and evaluates cutting-edge cell immunotherapies in model systems and patient samples at the interface of fundamental biology and cellular engineering with the ultimate goal of developing translatable insights to improve outcomes for brain tumor patients. In addition to the opportunity to contribute to this mission, this role offers the chance to present at laboratory meetings and departmental seminars, and dedicated prior technicians have presented their work at national scientific conferences and co-authored publications. Enthusiasm, work ethic and eagerness to contribute as part of a larger team are valued over specific skills; however, a documented record of at least two years (post-undergraduate) in an academic or industry laboratory setting is required. Qualifications

  • Generate and maintain CAR-T cells, including production of viral stocks, collection of primary T cells, cell transduction and expansion, and maintenance of associated records
  • Cell culture of diverse human cell lines, including brain tumor models and patient-derived organoids
  • Assay the activity of CAR-T cells in vitro, including cytokine generation, cytotoxicity, and cellular imaging
  • Flow cytometric analysis of CAR-T cells
  • Molecular cloning, including coordination and assessment of sequencing results and maintenance of documented records of constructs
  • Execution of protocol-based genomics assays including single cell sequencing, spatial transcriptomics, and spatial proteomics
  • Interface with collaborators and service core providers to transfer specimens, submit service requests, and oversee receipt of data and services. This may involve travel to local MGB hospitals to collect patient tissue specimens (shuttles available at no cost).
  • Basic laboratory management including inventory management, package receipt/shipping, routine instrument monitoring and maintenance, maintaining and distributing key lab protocols.
  • This role requires being listed as an emergency contact alongside other team members for mission-critical instrumentation (such as scientific freezers). In the unexpected event that such instrumentation fails, this may result in after-hours contact and a responsibility to help coordinate a response alongside other team members.
  • Attendance and presentation at weekly lab scientific meetings.

  • Experience with CAR-T cell production and relevant in vitro assays
  • Experience with molecular cloning, including validation of generated constructs
  • Prior mammalian cell culture work, including lentiviral production
  • Previous work performing single cell sequencing, spatial transcriptomics, or immunofluorescence microscopy
  • Experience with histology, including specimen preparation, cryosectioning or FFPE sectioning, and immunostaining
  • Software: Microsoft suite including Excel, Graphpad Prism, Flowjo, Snapgene or other molecular cloning software
